RINGKASAN


Penelitian ini bertujuan membuat vaksin polivalen dan vaksin polivalen plus Aeromonas hydrophila dengan penambahan vitamin C dan adjuvant serta mengetahui efikasi vaksin tersebut dalam mengendalikan penyakit MAS pada lele dumbo. Penelitian dilaksanakan menggunakan Rancangan Acak Lengkap (RAL) pola factorial dengan 2 variabel, 2 kali ulangan. Variabel terdiri atas perlakuan A berupa jenis vaksin, yaitu A1 = Vaksin polivalen, A2 = Vaksin polivalen dengan Vit C dan adjuvant alumunium potassium sulfat, A3 = Vaksin polivalen dengan Vit C dan adjuvant alumunium hidroksida, dan A4 = Kontrol (vaksinasi dengan PBS pH 7,0), serta perlakuan B berupa cara vaksinasi, yaitu B1 = suntik intraperitoneal, B2 = suntik intramuskular, dan B1 = rendaman. Lele dumbo yang digunakan berukuran panjang 10-13 cm dengan berat 19,1-26,7 g. Pada tiap perlakuan digunakan 8 ekor lele dumbo. Vaksinasi suntik diberikan dengan dosis 0,1 ml/ikan dengan kepadatan bakteri 107 sel/ikan, sedangkan vaksinasi rendaman dilakukan dengan cara merendam ikan uji dalam emulsi vaksin dengan konsentrasi 107 sel/ml selama 30 menit. Parameter yang diamati adalah titer antibodi, sintasan, Relative Percent Survival (RPS), Rerata Waktu Kematian (RWK) dan kualitas air. Analisis data menggunakan Analysis of Variance (ANOVA) dan uji lanjut dengan Duncans Multiple Range Test (DMRT). Booster dilakukan satu minggu setelah vaksinasi dan uji tantang dilakukan dua minggu setelah booster. Hasil penelitian menunjukkan bahwa vaksinasi dengan vaksin polivalen dan vaksin polivalen plus sel A. hydrophila dapat meningkatkan sintasan, RPS, dan titer antibodi (P<0.05). Vaksinasi secara suntik intraperitoneal, suntik intramuskular, maupun rendaman dapat menjadi alternatif pilihan untuk vaksinasi, karena ketiganya memiliki efektivitas yang sama dalam memberikan perlindungan relatif pada lele dumbo.

SUMMARY


The aims of this research were to make polyvalen and plus polyvalen of A. Hydrophila vaccines and to evaluate of the efficacy of these vaccines to control MAS disease on catfish. The research was constructed as factorial of CRD with two variables and twice replication. First variable were vaccine, i.e. A1 : polyvalen of vaccine, A2 : polivalen of vaccine with vitamine C dan alumunium potassium sulfat of adjuvant, A3 : polivalen of vaccine with vitamine C dan alumunium hidroksida of adjuvant, dan A4 : control (vaccination with PBS pH 7,0), and second variables were vaccination, i.e. B1 : intraperitoneal of injection vaccination, B2 : intramuskular of injection vaccination, and B3 : immersion vaccination. The research was used catfish with length 10-13 cm and 19.1-26.7 g of weight and a number of 8 fish into one of treatment. Injection vaccination was conducted with 0.1 ml dosage vaccine at 107 cell/fish. Immersion vaccination was done by immersing the fishes at 107 cell/fish for 30 minutes. Booster was conducted one week after vaccination and challenge test was conducted two weeks after booster. Parameter to controlled were titer antibody, survival rate, Relative Percent Survival/RPS, to death (MTD) and water quality. Futhermore, they were analized by Analysis of Variance and Duncans Multiple Range Test (DMRT). The result showed that polyvalen and plus polyvalen of A. hydrophila vaccines could increase RPS, survival rate, and the titer of antibody (P<0.05). Intraperitoneal of injection vaccination, intramuskular of injection vaccination, and immersion vaccination were the effective vaccination could protect to catfish.
