Several assays have been applied for estrogenic activity determination. In vivo assays employed animal models is unsuitable for large-scale screening due to its time and cost consuming. Thus, in vitro assays become more widely used. Examples of in vitro assays are competitive ligand binding assays, cell proliferation assays, recombinant receptor gene assay in human cells and Yeast Estrogen Screen (YES) method. In vitro assays are useful for screening and assessing estrogenic activity of many xenoestrogens and phytoestrogens. Understanding the principle, advantages and limitation of each assay, as well as attempts to overcome its limitation, might help for applying the best assay for estrogenic activity determination on particular estrogenic compounds. This paper provides discussion on the principle, advantages and limitation of in vitro bioassays for estrogenic activity determination and recent development of each assay particularly in YES method.
