INFLUENCE OF 2,4-D (DICLOROPHENOXY ACETIC ACID) AND BAP (BENZYL AMINO PURINE) ON CALLUS PROLIFERATION AND SECONDARY METABOLITE PRODUCTION FROM KAEMFERIA GALANGA CALLUS

The aim of this research is to find combination of 2,4-D (diclorophenoxy acetic acid) and BAP (benzyl amino purine) concentration which give the best influence to callus proliferation and to know the effect of interaction between 2,4-D (diclorophenoxy acetic acid) and BAP (benzyl amino Purine) to obtain good growth culture of Kaemferia galanga L callus and able to produce secondary metabolite. The study was conducted at Tissue Culture Laboratory, Integrated Laboratory University of Muhammadiyah Purwokerto, the research time was held for 5 months. The design used for callus proliferation was a combination of 2.4 D (0.5 - 2 ppm) and BAP (0 - 0.2 ppm). The combination of effective treatment for callus proliferation was 2.4 D concentrations of 1 to 3 ppm and without the addition of BAP (B0) which showed the result of callus proliferation having callus volume, fresh callus weight, callus dry weight and callus morphology callus ( colour and texture callus) was better than the addition of BAP, however 2.4 D with 1 ppm concentration gave the best callus proliferation rate indicated at callus volume of 1.83 cm³, fresh weight of callus weighing 1,793 g, 0.077 g of dried callus weight and the weakness high and white light colour with friable nature, while the higher level of 2,4 D given effect on callus color formation to green and lead to organogenesis process (shoot and root formation). Based on qualitative analysis test using thin layer chromatography (TLC ), Extract methanol callus kencur research results contain secondary metabolite compounds in the form of alkaloids, flavonoids, Tannins, saponins, steroids and ethyl para-methoxycinnamate.
