IDENTIFICATION OF DOG FOR HALAL AUTHENTIFICATION WITH GAS CHROMATOGRAPHY MASS SPECTROSCOPY (GCMS) AND CHEMOMETRICS

Due to the wide difference of meat price, some unethical seller replaces beef intentionally with dog meat to get economical profits. The objective of this study is to assess the suitability of Gas Chromatography Mass Spectrophotometri coupled with chemometrics of partial least square (PLS) and principal component analysis (PCA) for rapid screening of dog fatt in beef meatball formulation. The lipid fraction of meatball is obtained by employing Bligh and Dyer extraction method using chloroform:methanol (1:2)/water as extracting solvents. The GCMS chromatogram correlated with composition of triacylglyserols from beef, dog, chicken, pork and rat fat were scanned, interpreted, and identified. Qualitative differences among GCMS chromatogram of TAG are proposed as a basis tools for differentiating between dog fat and other fats. The main TAG composing dog are lauric acid (0.47%), pentadecanoic acid (0.16%), myristic acid (2.66%), 9-octadecenoic acid (37.71%), myristoleic acid (0.15%), palmitoleic acid (0.36%), palmitic acid (22.90%), palmitoleic acid (5.70%), stearic acid (9.03%), heptadecanoic acid (0.43%), cis 10 heptadecanoic acid (0.17%), linoleic acid (17.73%), nonadecanoic acid (0.10%), arachidonic acid (0.36%), 6,9,12-octadecatrienoic acid (0.15%), 11, eicosandienoic acid (0.94%), eicosapentanoic acid (0.36%), methyl gamma linolenate (0.23%). PCA from GCMS chromatogram was capable to identified fat from dog, chicken , beef, pork and goat. Using score plot of PCA, dog has similiarity with chicken and pork.
